Glabridin is a flavonoid found in the root extract of Glycyrrhiza glabra (licorice). Its use in acne management is supported by a modest but growing body of scientific evidence, primarily from in vitro and small-scale clinical studies. Glabridin exhibits notable anti-inflammatory and antioxidant properties, which are relevant to acne pathogenesis, as inflammation and oxidative stress play crucial roles in acne development and exacerbation. Several studies have demonstrated that glabridin can inhibit the activity of cyclooxygenase and reduce the production of pro-inflammatory cytokines, thereby helping to diminish skin inflammation.

Additionally, glabridin has been shown to possess mild antibacterial activity against Propionibacterium acnes (now Cutibacterium acnes), the bacteria implicated in acne. It also inhibits melanin synthesis, which may help reduce post-inflammatory hyperpigmentation associated with acne lesions. However, most of these findings are based on laboratory models or topical formulations tested in small human trials, often as part of multi-ingredient products. There is a lack of large, well-controlled clinical trials specifically evaluating glabridin’s effectiveness as a monotherapy for acne.

In conclusion, while there is preliminary scientific support for the use of glabridin in acne due to its anti-inflammatory and antibacterial properties, the current evidence base is limited and does not yet justify a strong recommendation. Further research, especially clinical studies, is needed to fully establish its efficacy and safety.
