Glycyrrhizic acid, a main bioactive compound from licorice root (Glycyrrhiza glabra), has a long history of traditional use in herbal medicine to support the adrenal glands and address symptoms commonly associated with adrenal fatigue, such as fatigue and stress. This traditional use is rooted in the herb’s adaptogenic reputation in systems like Traditional Chinese Medicine and Ayurveda, where licorice is believed to "tonify" or restore balance to the body's stress response. The scientific basis for its direct impact on adrenal gland function is limited. Some research suggests glycyrrhizic acid inhibits 11β-hydroxysteroid dehydrogenase type 2 (11β-HSD2), an enzyme that converts active cortisol to inactive cortisone, thereby increasing cortisol’s half-life in the body. This could theoretically support adrenal function in cases of low cortisol, but it also raises the risk of pseudoaldosteronism, a condition characterized by hypertension, edema, and low potassium. While these mechanisms have been documented, robust clinical trials directly showing glycyrrhizic acid benefits for adrenal health are lacking. Therefore, while there is a strong tradition for its use in adrenal support, scientific evidence remains limited and inconsistent.
