Glycyrrhizin, a major active compound found in licorice root (Glycyrrhiza glabra), has a long history of traditional use for various ailments, including as an adaptogen for adrenal support. From a scientific perspective, glycyrrhizin inhibits the enzyme 11β-hydroxysteroid dehydrogenase type 2 (11β-HSD2), which normally converts active cortisol into inactive cortisone in the kidneys. By inhibiting this enzyme, glycyrrhizin can indirectly increase the availability of cortisol in the body.

This mechanism is why glycyrrhizin has been proposed to support adrenal function, particularly in cases of "adrenal fatigue" or suboptimal cortisol production. Some small clinical studies and case reports suggest that licorice extracts may help maintain cortisol levels in patients with Addison's disease (adrenal insufficiency), allowing for lower doses of corticosteroid medications. However, the direct evidence in healthy individuals or for general adrenal "support" is limited.

Caution is necessary, as glycyrrhizin can cause significant side effects—such as hypertension, hypokalemia, and sodium retention—due to its effects on mineralocorticoid receptors, especially with high doses or prolonged use. Overall, the scientific evidence supporting glycyrrhizin for adrenal support is moderate (rated 3/5), with its use better established in specific medical contexts rather than for general wellness.
