Vitamin B5, also known as pantothenic acid, has a scientific basis for its role in supporting the adrenal glands, though the evidence is limited and primarily preclinical. Pantothenic acid is essential for the synthesis of coenzyme A (CoA), which is crucial in numerous metabolic pathways, including the synthesis of adrenal hormones such as cortisol. Animal studies, particularly in rats, have shown that pantothenic acid deficiency can lead to impaired adrenal function and reduced synthesis of adrenal hormones. Some older studies, such as those from the mid-20th century, observed adrenal atrophy in animals subjected to pantothenic acid deficiency.
However, robust human clinical trials demonstrating a direct benefit of supplemental vitamin B5 for adrenal gland function or for so-called "adrenal fatigue" are lacking. Most evidence in humans is extrapolated from the known biochemical roles of B5 and case reports rather than controlled studies. The rationale for use in supplements typically hinges on the vitamin's role in hormone synthesis rather than on clinical outcomes.
