Vitamin B5, also known as pantothenic acid, plays a key role in the synthesis of coenzyme A, which is essential for energy metabolism and the synthesis of steroid hormones, including those produced by the adrenal glands. The rationale for its use in supporting adrenal function primarily stems from its biochemical role: coenzyme A is involved in the adrenal cortex’s production of cortisol and other hormones. Some animal studies from the mid-20th century suggested that pantothenic acid deficiency led to impaired adrenal function and reduced hormone production. These findings have been extrapolated to humans, and vitamin B5 is sometimes recommended in “adrenal support” supplements, especially in the context of stress or fatigue.

However, direct human evidence to support the use of vitamin B5 supplementation specifically for adrenal support is limited and generally of low quality. There are few, if any, well-controlled clinical trials demonstrating that supplementation improves adrenal gland function in healthy individuals or those with stress-related conditions. Most people consuming a balanced diet are unlikely to be deficient in vitamin B5, as it is widely available in foods. Thus, while there is a plausible biochemical mechanism and some historical animal data, the scientific evidence in humans is weak, and routine supplementation for adrenal support is not strongly validated by current research.
