Pantethine, a derivative of vitamin B5 (pantothenic acid), is sometimes used in traditional and naturopathic medicine to support adrenal gland function. This practice is based primarily on the theoretical role of pantothenic acid in the biosynthesis of coenzyme A, which is necessary for the production of adrenal hormones such as cortisol. The adrenal glands require coenzyme A for steroidogenesis, and animal studies from the mid-20th century showed that pantothenic acid deficiency could impair adrenal function. Historically, pantothenic acid has been associated with general stress support and adrenal "health" in alternative medicine literature, but these uses are rooted in tradition rather than robust clinical research.

However, there is limited direct scientific evidence from human studies to confirm that pantethine or pantothenic acid supplementation has a significant or measurable effect on adrenal health or hormone production in otherwise healthy individuals. Most modern research on pantethine focuses on its potential lipid-lowering effects rather than adrenal support. While the biochemical rationale is plausible, clinical trials specifically assessing the impact of pantethine on adrenal function are lacking. Thus, while use of pantethine for adrenal support is common in traditional and naturopathic circles, this use is not strongly validated by high-quality scientific studies.
