Bioflavonoids, a diverse group of plant-derived polyphenolic compounds, have been investigated for their potential neuroprotective effects in Alzheimer's Disease (AD). Preclinical studies, primarily in vitro and in animal models, suggest that certain bioflavonoids (such as quercetin, rutin, and hesperidin) may reduce oxidative stress, decrease neuroinflammation, inhibit amyloid-beta aggregation, and improve cognitive function. These mechanisms are relevant because oxidative stress and amyloid plaque formation are key pathological features of AD. Some studies have also found that diets rich in flavonoids are associated with a lower risk of cognitive decline in epidemiological cohorts.

However, clinical evidence in humans is limited. Randomized controlled trials specifically testing bioflavonoids for AD treatment are scarce, and those that exist are generally small, short-term, and often inconclusive. The bioavailability and metabolism of bioflavonoids in humans also present challenges, as it is unclear if sufficient active compounds reach the brain at therapeutic levels. Thus, while there is a plausible scientific rationale and some early supportive data, the overall quality and quantity of evidence supporting the use of bioflavonoids for Alzheimer's Disease is modest, and major clinical guidelines do not recommend their use as a standard therapy. More robust human trials are needed to establish efficacy and safety.
