Synopsis

Source of validity: Scientific
Rating (out of 5): 2

Epigallocatechin gallate (EGCG), a major polyphenol in green tea, has been investigated for its potential neuroprotective effects in Alzheimer's Disease (AD). Preclinical studies in cell cultures and animal models suggest that EGCG may reduce amyloid-beta aggregation, inhibit tau phosphorylation, and modulate oxidative stress and neuroinflammation—key pathological features of AD. For example, in vitro studies demonstrate that EGCG can destabilize amyloid-beta fibrils and reduce their neurotoxicity. Animal studies have also reported improvements in cognitive function and reductions in AD pathology following EGCG administration.

However, human clinical evidence remains limited and inconclusive. Few clinical trials have tested EGCG directly in patients with Alzheimer's Disease, and existing studies often have small sample sizes, short durations, or use green tea extracts rather than purified EGCG. Most human studies focus on cognitive function in healthy adults or those with mild cognitive impairment, with mixed results. Systematic reviews and meta-analyses conclude that while preclinical data are promising, there is insufficient high-quality clinical evidence to recommend EGCG as a treatment or preventative strategy for Alzheimer's Disease at this time.

In summary, EGCG's use in Alzheimer's Disease is supported by scientific rationale and encouraging preclinical results, but its clinical efficacy and safety remain to be definitively established in humans. The current evidence base rates as low to moderate (2/5).
