Ginkgoflavones are a group of flavonoid compounds found in the leaves of the Ginkgo biloba tree. Extracts from Ginkgo biloba, which contain ginkgoflavones as well as terpene lactones, have been traditionally used in Chinese medicine to support cognitive health. In modern research, Ginkgo biloba extract (often standardized to contain 24% flavone glycosides, including ginkgoflavones) has been studied for its potential benefits in Alzheimer’s Disease (AD) and other forms of dementia. The rationale is based on purported antioxidant, anti-inflammatory, and neuroprotective effects, as well as improvements in cerebral blood flow. However, clinical evidence for the efficacy of ginkgoflavones or Ginkgo biloba extract in Alzheimer’s Disease is mixed. Some randomized controlled trials and systematic reviews have shown modest benefits in cognitive function and activities of daily living in patients with mild to moderate dementia, but the effect sizes are small and clinical relevance is debated. The largest and most rigorous trials (such as the GEM and GuidAge studies) did not find significant prevention or slowing of dementia onset. Major guidelines such as those from the American Academy of Neurology and the National Institute for Health and Care Excellence do not recommend Ginkgo biloba extract as a standard treatment for Alzheimer’s Disease due to insufficient and inconsistent evidence. In summary, while there is some scientific investigation and limited support for ginkgoflavones in cognitive decline, the overall evidence is weak and not sufficient for strong clinical recommendations.
