Growth factors have been investigated as potential therapeutic agents for Alzheimer's Disease (AD) due to their roles in neuronal survival, differentiation, and synaptic plasticity. Scientific research, primarily in preclinical models and early-phase human trials, has focused on growth factors such as nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF), and insulin-like growth factor 1 (IGF-1). These molecules are critical for the maintenance of neuronal health, and decreased levels have been observed in the brains of AD patients.

Animal studies have demonstrated that administering certain growth factors can reduce neurodegeneration, improve cognitive function, and decrease hallmark AD pathologies such as beta-amyloid deposition and tau hyperphosphorylation. For example, NGF delivery to the basal forebrain in animal models has shown neuroprotective effects. Some early-phase clinical trials, including gene therapy approaches delivering NGF, have suggested possible benefits in slowing cognitive decline, but these studies have been small and results remain inconclusive.

Despite promising preclinical data, translating these findings into effective human treatments has proven challenging. Issues include difficulties in delivering growth factors across the blood-brain barrier, potential side effects, and lack of robust large-scale clinical trial data demonstrating clear clinical benefit. Thus, while there is a scientific rationale and some early evidence, the overall level of clinical evidence supporting the use of growth factors in treating Alzheimer's Disease is currently limited, meriting a moderate evidence rating.
