Hesperetin is a citrus flavonoid primarily found in oranges and other citrus fruits. Its use in the context of Alzheimer's Disease (AD) is grounded in preclinical scientific research rather than traditional medicine. Several in vitro and animal studies have investigated hesperetin's neuroprotective effects, attributing its benefits to antioxidant, anti-inflammatory, and anti-amyloidogenic properties. For example, experimental models have shown that hesperetin can reduce oxidative stress, inhibit neuroinflammation, and attenuate amyloid-beta (Aβ) aggregation and toxicity—hallmarks of Alzheimer's pathology. Further, hesperetin has demonstrated the ability to modulate signaling pathways such as the Nrf2/ARE pathway, which plays a role in cellular defense mechanisms against oxidative damage. Some studies also suggest hesperetin may improve cognitive function in animal models of AD. However, direct evidence from human clinical trials is lacking. Most findings are limited to laboratory or animal research, and the doses used in these studies are often much higher than what could be achieved through diet or supplementation in humans. There is currently insufficient evidence to recommend hesperetin as a treatment for Alzheimer's Disease in clinical practice. More research, particularly well-designed human studies, is needed to confirm its efficacy and safety in this context.
