Iridoids are a class of secondary plant metabolites found in various medicinal plants such as Gardenia jasminoides, Gentiana species, and Rehmannia glutinosa. There is a growing body of preclinical scientific evidence suggesting that iridoids may have neuroprotective effects relevant to Alzheimer's Disease (AD). Laboratory studies (in vitro and in animal models) indicate that certain iridoids can inhibit amyloid-beta aggregation, reduce neuroinflammation, and ameliorate oxidative stress—all of which are hallmarks of AD pathology. For example, geniposide (an iridoid glycoside from Gardenia jasminoides) has been shown to protect neuronal cells against amyloid-beta-induced toxicity and improve cognitive performance in animal models of AD. Other iridoids like catalpol (from Rehmannia glutinosa) have demonstrated similar effects, including enhancement of synaptic plasticity and reduction of tau hyperphosphorylation in preclinical studies.

However, evidence in humans remains extremely limited. There are no large-scale, high-quality clinical trials directly evaluating iridoids for the prevention or treatment of AD in people. Most available data are derived from cell-based assays and rodent models, and while these findings are promising, they do not constitute definitive proof of efficacy in clinical settings. Thus, scientific validation exists at the preclinical level, but clinical evidence is lacking, resulting in a moderate evidence rating of 2 out of 5.
