Isoliquiritigenin is a flavonoid compound found primarily in licorice roots (Glycyrrhiza uralensis) and has attracted attention for its neuroprotective and anti-inflammatory properties. Scientific interest in isoliquiritigenin as a potential agent for supporting or treating Alzheimer's Disease (AD) stems mainly from in vitro and animal model studies published over the last decade. Research has demonstrated that isoliquiritigenin can inhibit acetylcholinesterase activity, reduce neuroinflammation, and attenuate oxidative stress—mechanisms relevant to the pathology of Alzheimer's Disease. For example, preclinical studies report that isoliquiritigenin reduces amyloid-beta-induced neurotoxicity and suppresses microglial activation in mouse models, which are key features of AD progression. Moreover, some studies suggest it improves cognitive performance in rodent models of Alzheimer-like conditions. However, there is a lack of clinical trials in humans evaluating the efficacy and safety of isoliquiritigenin specifically for Alzheimer's Disease. The majority of supporting data is preclinical, with only a few studies directly addressing cognitive outcomes. Thus, while the biological plausibility and animal data provide a scientific basis for further investigation, the current level of evidence is limited and does not justify clinical use at this time. More well-designed human trials are necessary to determine its therapeutic potential and safety profile in Alzheimer's patients.
