Nicotinamide Mononucleotide (NMN) is a precursor to NAD+ (nicotinamide adenine dinucleotide), a molecule essential for cellular energy metabolism and DNA repair. In Alzheimer’s disease (AD), NAD+ levels are often reduced, and this decline has been implicated in neurodegeneration. Several preclinical studies—mainly in animal models—suggest that NMN supplementation may improve cognitive function, enhance neuronal survival, and reduce neuroinflammation by restoring NAD+ levels and supporting mitochondrial function. For example, mouse models of AD have shown improvements in memory and synaptic plasticity following NMN administration. Mechanistically, NMN may activate sirtuins and other NAD+-dependent enzymes that protect against oxidative stress and amyloid-beta toxicity, both hallmarks of AD pathology.
However, human data are lacking. To date, no large-scale clinical trials have confirmed the efficacy or safety of NMN in Alzheimer’s patients. Most available evidence comes from animal studies or small pilot human studies focused on general aging or metabolic health, not specifically Alzheimer’s disease. Therefore, while the scientific rationale is plausible and early results are encouraging, robust clinical validation is currently insufficient. NMN is not an approved treatment for Alzheimer’s, and its use for this condition should be considered experimental.
