Omega-3 fatty acids, particularly docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA), have attracted scientific interest for their potential role in supporting cognitive function and slowing the progression of Alzheimer's Disease (AD). These fatty acids are primary components of neuronal cell membranes and play important roles in anti-inflammatory and neuroprotective processes. Epidemiological studies have indicated that populations with higher dietary intake of omega-3s, especially from fish, have lower rates of cognitive decline and dementia. Preclinical studies in animal models have shown that omega-3s can reduce amyloid-beta deposition and neuroinflammation, which are hallmarks of AD pathology. However, clinical trials in humans have produced mixed results. Some small studies and observational data suggest a modest benefit in slowing cognitive decline in individuals with mild cognitive impairment or early Alzheimer's Disease. Large randomized controlled trials, such as the MAPT and OmegAD studies, have generally failed to demonstrate significant cognitive improvement in patients with established AD, though some subgroups (such as those with low baseline omega-3 levels or certain genetic backgrounds) may benefit. Overall, there is a scientific rationale for omega-3 supplementation in the context of AD prevention or early intervention, but current evidence from clinical trials does not robustly support its use as a treatment for established Alzheimer's Disease. More research is needed to clarify which populations, if any, might benefit most from omega-3 supplementation.
