There is some scientific interest in the use of Vitamin B3 (niacinamide) for Alzheimer's Disease, but current evidence is limited and preliminary. Laboratory and animal studies have suggested that niacinamide may have neuroprotective effects, possibly by inhibiting tau phosphorylation, improving mitochondrial function, and reducing oxidative stress in neural tissue. A notable preclinical study published in "The Journal of Neuroscience" (2008) found that high doses of niacinamide improved cognitive function and reduced tau pathology in a mouse model of Alzheimer's Disease. However, human clinical trials are lacking. Only a few small-scale or early-phase studies have investigated niacinamide in Alzheimer's patients, and robust, large-scale, placebo-controlled trials have not been completed. The available human data do not provide convincing evidence to recommend niacinamide as a treatment for Alzheimer's Disease. Major guidelines and reviews, including those from the Alzheimer’s Association and Cochrane Reviews, do not endorse niacinamide for this indication due to insufficient clinical evidence. In summary, while there are plausible mechanisms and encouraging preclinical findings, the current scientific validation is weak (rated 2/5), and niacinamide should not be considered a proven or established therapy for Alzheimer's Disease at this time. Further research, particularly well-designed human studies, is needed to determine its efficacy and safety for this condition.
