Vitamin B3 refers to a group of compounds including niacin (nicotinic acid) and nicotinamide (niacinamide). There is some scientific interest in the potential of vitamin B3 derivatives, particularly nicotinamide and nicotinamide riboside, for supporting or treating Alzheimer's Disease (AD), but the evidence is currently limited and largely preclinical. Animal studies have suggested that high doses of nicotinamide may reduce tau phosphorylation and improve cognitive performance in mouse models of AD. For example, a widely cited 2008 study found that nicotinamide administration improved memory and reduced pathological tau in transgenic mice. More recently, research has explored how boosting NAD+ (a metabolite related to B3) via precursors like nicotinamide riboside may affect neurodegeneration, with some promising findings in cellular and animal models. However, human clinical trials are very limited. A few small pilot studies have examined nicotinamide’s safety and potential cognitive benefits in mild cognitive impairment and AD, but results are inconclusive and sometimes show no significant effect. There is no robust evidence from large, well-controlled human trials to support routine use of vitamin B3 for Alzheimer’s prevention or treatment at this time. Nevertheless, the biological rationale is being actively explored, as vitamin B3 is involved in pathways relevant to neuronal health and aging. In summary, there is emerging, but still insufficient, scientific evidence for vitamin B3’s role in Alzheimer’s Disease.
