Some clinical trials and observational studies have suggested that vitamin D supplementation may improve menstrual regularity and ovulatory function in women with amenorrhea related to PCOS or hypovitaminosis D, either alone or in combination with other treatments such as metformin or calcium. For example, a 2015 systematic review found modest improvements in menstrual frequency in women with PCOS after vitamin D supplementation, although evidence quality was low and results were inconsistent. No large-scale, high-quality randomized controlled trials have established vitamin D as a standalone treatment for amenorrhea, and its effectiveness appears limited to subgroups with underlying deficiency.
