Aniracetam is a nootropic compound belonging to the racetam family. Its use for supporting or treating anxiety is primarily justified by preclinical scientific research, rather than tradition or widespread clinical validation. Animal studies—particularly in rodents—have demonstrated that aniracetam may exert anxiolytic (anxiety-reducing) effects. These effects are thought to be mediated by modulation of glutamatergic and cholinergic neurotransmission, and possibly through interactions with AMPA receptors and dopaminergic pathways in the brain. For example, a study by Nakamura and colleagues (2001) found that aniracetam reduced anxiety-like behavior in mice in the elevated plus-maze test. Another study (Nakamura & Kurasawa, 2001) suggested that metabolites of aniracetam contributed to its anxiolytic action. However, evidence in humans is limited. There are a few small-scale, preliminary clinical trials and anecdotal reports suggesting potential benefits for anxiety, but no large, high-quality randomized controlled trials have confirmed these effects. As such, while the preclinical scientific evidence is promising and provides a plausible mechanism of action, the overall evidence base is limited, and clinical recommendation for treating anxiety in humans with aniracetam is not currently supported by robust scientific consensus.
