Glycine, a non-essential amino acid, has some scientific evidence supporting its use for anxiety, although the evidence is limited and not yet robust. Glycine acts as an inhibitory neurotransmitter in the central nervous system, particularly in the spinal cord, brainstem, and retina, and also modulates NMDA (N-methyl-D-aspartate) receptors in the brain. Some preclinical studies suggest that glycine supplementation can have anxiolytic (anxiety-reducing) effects, potentially by enhancing inhibitory neurotransmission and reducing overexcitation in neural circuits associated with anxiety.
A few small human studies and case reports have explored glycine’s role in sleep improvement and reduction of subjective stress, which relates indirectly to anxiety. For example, a randomized controlled trial found that glycine improved subjective sleep quality and reduced fatigue in individuals with sleep complaints—a common comorbidity of anxiety disorders. However, direct evidence from large, well-designed clinical trials specifically targeting anxiety as a primary outcome is lacking.
While animal studies and mechanistic research provide a rationale for further investigation, current human data are insufficient to recommend glycine as a primary treatment for anxiety. Its use is not based on traditional medicine but rather on emerging scientific understanding of its neurophysiological roles. Further research is needed to clarify its efficacy and safety profile for this indication.
