7,8-Dihydroxyflavone (7,8-DHF) is a small-molecule flavonoid that has attracted attention in neuroscience due to its action as a selective agonist of the TrkB receptor, which is the primary receptor for brain-derived neurotrophic factor (BDNF). Dysregulation of BDNF-TrkB signaling has been implicated in anxiety and mood disorders. Several preclinical studies (in animal models) have shown that 7,8-DHF can produce anxiolytic-like effects, presumably by enhancing TrkB signaling in brain regions involved in anxiety regulation (such as the hippocampus and amygdala). For example, Science Translational Medicine (2010) and Neuropsychopharmacology (2012) published studies showing reduced anxiety-like behaviors in mice after administration of 7,8-DHF.

However, evidence in humans is lacking. To date, no clinical trials have been completed to evaluate the efficacy or safety of 7,8-DHF for anxiety disorders in people. As such, its use is not supported by clinical guidelines or mainstream psychiatric practice. Most of the evidence is limited to the preclinical (animal) level, and results may not translate directly to human anxiety disorders.

In summary, while there is some scientific rationale and preclinical evidence suggesting a potential role of 7,8-DHF in modulating anxiety-like behaviors, the overall evidence is weak (rating: 2/5) due to the absence of human studies.
