Apigenin is a naturally occurring flavonoid found in various plants such as chamomile, parsley, and celery. Its traditional use for anxiety is primarily linked to chamomile, which has a long history in herbal medicine for calming effects. However, the evidence for apigenin specifically as an anxiolytic comes from preclinical scientific research. Animal studies have demonstrated that apigenin can bind to benzodiazepine receptors on the GABAA receptor complex in the brain, which is the same mechanism as many pharmaceutical anxiolytics. These studies report reduced anxiety-like behavior in rodents after apigenin administration. Additionally, in vitro work supports its modulatory role on GABAergic neurotransmission, which is central to anxiety regulation. Despite these promising results, human clinical data are limited. Most human studies have investigated chamomile extracts rather than isolated apigenin, and while some small trials suggest anxiolytic benefits of chamomile, it is unclear how much apigenin specifically contributes. There are no large-scale, high-quality clinical trials of isolated apigenin for anxiety disorders in humans to date. Therefore, while there is a scientific rationale and preliminary evidence from animal studies, the direct evidence in humans is weak, justifying a rating of 2.
