Griffonia simplicifolia is a West African plant whose seeds are a natural source of 5-hydroxytryptophan (5-HTP), a direct precursor to serotonin. Serotonin is a neurotransmitter involved in mood regulation, and low serotonin levels are associated with anxiety disorders. The rationale for using Griffonia simplicifolia in anxiety is thus based on increasing brain serotonin levels via 5-HTP supplementation. Several small-scale human and animal studies suggest that 5-HTP supplementation may have anxiolytic (anxiety-reducing) effects. For example, some clinical trials have shown that 5-HTP can reduce symptoms in people with anxiety or panic disorders, but the number of well-designed, large randomized controlled trials (RCTs) is limited. Most available studies are either small, short-term, or focus on depressive symptoms rather than anxiety specifically. Systematic reviews generally conclude that while there is biological plausibility and preliminary evidence, the clinical evidence is insufficient to strongly recommend 5-HTP (or Griffonia simplicifolia) for anxiety disorders. Safety is another consideration, as 5-HTP can cause side effects and interact with medications.
