Pyridoxine (Vitamin B6) has been investigated for its potential role in supporting or treating anxiety disorders, primarily due to its involvement in neurotransmitter synthesis. Vitamin B6 acts as a cofactor in the biosynthesis of gamma-aminobutyric acid (GABA), serotonin, and dopamine—neurotransmitters implicated in mood regulation and anxiety. Several small clinical studies and observational data have explored the relationship between B6 status and anxiety symptoms. For example, a 2022 randomized, placebo-controlled trial found that high-dose B6 supplementation modestly reduced self-reported anxiety levels in young adults, possibly through enhanced GABAergic activity, though the effect size was small and the study duration short.

Despite these findings, the overall scientific evidence remains limited and inconsistent. Most studies to date are small, use self-reported outcomes, and do not establish causality. Meta-analyses and systematic reviews generally conclude that while there is biological plausibility for B6’s involvement in anxiety pathways, robust clinical evidence is lacking, and supplementation is not a standard or widely recommended treatment for anxiety disorders. Current guidelines do not recommend B6 as a primary or adjunctive therapy for anxiety. Thus, while scientific rationale exists and preliminary data are somewhat promising, the strength of evidence is modest at best, warranting a rating of 2.
