Valerenic acid is a key component of Valeriana officinalis (valerian) root, a traditional herbal remedy for anxiety and insomnia. Scientific interest in valerenic acid derivatives arose from their activity on the central nervous system, particularly their modulation of GABAA receptors. Several preclinical studies in animal models have shown that valerenic acid can bind to GABAA receptors and enhance inhibitory neurotransmission, which is a known mechanism targeted by many anxiolytic drugs (e.g., benzodiazepines).
Despite promising preclinical results, high-quality human clinical studies are limited. Some small randomized controlled trials of valerian extracts (which contain valerenic acid among other compounds) suggest mild anxiolytic effects, but results are inconsistent and often confounded by the presence of other active constituents. Furthermore, the specific contribution of valerenic acid derivatives, as opposed to whole valerian extract, in treating anxiety disorders has not been conclusively demonstrated. Systematic reviews and meta-analyses typically rate the evidence for valerian's efficacy in anxiety as low to moderate, citing methodological limitations and variability in preparations.
