Mevinolin, also known as lovastatin, is a statin medication that is scientifically validated for use in the treatment and prevention of arteriosclerosis (also referred to as atherosclerosis). Statins, including mevinolin, work by inhibiting the enzyme HMG-CoA reductase, which plays a crucial role in the biosynthesis of cholesterol in the liver. Elevated cholesterol, particularly low-density lipoprotein (LDL) cholesterol, is a well-established risk factor for the development of arteriosclerotic plaques in arterial walls.

Multiple large-scale, randomized controlled trials (RCTs) have demonstrated that statins significantly reduce LDL cholesterol levels and lower the risk of major cardiovascular events, such as myocardial infarction and stroke, in both primary and secondary prevention settings. The landmark Scandinavian Simvastatin Survival Study (4S) and subsequent studies like the West of Scotland Coronary Prevention Study (WOSCOPS) have firmly established the role of statins in slowing the progression of arteriosclerosis and reducing associated morbidity and mortality.

Guidelines from major cardiovascular societies, such as the American Heart Association (AHA) and the European Society of Cardiology (ESC), recommend statin therapy—including lovastatin—for patients at risk of, or diagnosed with, arteriosclerotic cardiovascular disease. Therefore, the use of mevinolin for arteriosclerosis is strongly backed by scientific evidence and is considered standard of care in medical practice.
