Pyridoxal-5-phosphate (P-5-P) is the active coenzyme form of vitamin B6, which plays a critical role in amino acid metabolism, glycogenolysis, and neurotransmitter synthesis. Its relevance to athletic performance and exercise stems from its involvement in the conversion of stored glycogen to glucose, facilitating energy production during physical activity, and in supporting protein metabolism, which is essential for muscle repair and growth.
Several studies have observed that intense exercise can increase the requirement for vitamin B6, as metabolic demand rises. Deficiency in vitamin B6 can impair physical performance and limit muscle function, but such deficiencies are rare in well-nourished individuals. Supplementation with P-5-P specifically, as opposed to other forms of vitamin B6, has not been robustly studied in athletes, and most trials focus on generalized vitamin B6 status or supplementation. No large, high-quality randomized controlled trials have demonstrated that P-5-P supplementation improves strength, endurance, or recovery in athletes who are not deficient in vitamin B6.
Therefore, while there is a sound biochemical rationale and some small-scale evidence for ensuring adequate B6 intake (and thus P-5-P levels) to support metabolism during exercise, there is limited direct scientific validation for using P-5-P as a specific ergogenic aid in healthy, well-nourished athletes. Supplementation may be beneficial only in cases of marginal or overt deficiency. Thus, the evidence is rated as 2 out of 5—suggesting a theoretical basis but lacking strong clinical validation for performance enhancement in the general athletic population.
