Vitamin B6 (pyridoxine) has been investigated as a potential treatment for autism spectrum disorder (ASD) since the 1960s. Early reports and small, uncontrolled studies suggested some individuals with autism might benefit from high-dose B6, particularly in combination with magnesium. The rationale was based on observations of altered B6 metabolism in some people with autism and its role as a cofactor in neurotransmitter synthesis. However, subsequent research has not consistently validated these early findings. Most controlled trials have shown little to no benefit for core autism symptoms, and systematic reviews, including those by the Cochrane Collaboration, conclude that there is insufficient evidence to support the use of vitamin B6 (with or without magnesium) for treating autism. While vitamin B6 supplementation is generally safe at recommended dosages, high doses may cause neuropathy and other side effects. The continued use of B6 for autism is primarily based on historical precedent and anecdotal reports, rather than robust, reproducible scientific validation. Current clinical guidelines do not recommend vitamin B6 as a standard treatment for autism spectrum disorder.
