Tylophorine is an alkaloid derived from the plant Tylophora indica, traditionally used in Ayurvedic medicine for respiratory and inflammatory conditions. Recent scientific investigations, primarily in vitro and in animal models, have suggested that tylophorine and its derivatives possess notable immunosuppressive and anti-inflammatory properties. Studies demonstrate that tylophorine can inhibit lymphocyte proliferation, suppress cytokine production (such as TNF-α and IL-2), and modulate the NF-κB pathway, which plays a pivotal role in immune response and inflammation. These mechanisms are relevant to autoimmune disorders, which are characterized by overactivity of the immune system. However, while these preclinical findings provide a plausible rationale for tylophorine’s use in autoimmune conditions, there is a lack of robust clinical trials in humans. Most evidence is limited to laboratory settings or animal studies, and the safety profile in humans, especially for long-term use, remains inadequately characterized. Thus, while there is some scientific basis for tylophorine’s immunomodulatory activity, the current level of evidence supporting its clinical use in autoimmune disorders is limited and insufficient for routine recommendation. More well-designed clinical studies are necessary to establish its efficacy and safety profile in humans.
