1-androsterone (also known as 1-androstene-3b-ol-17-one or 1-DHEA) is a synthetic prohormone that is sometimes marketed in bodybuilding supplements for its purported ability to increase muscle mass and strength by converting in the body to 1-testosterone (dihydroboldenone), a potent anabolic steroid. There is some scientific rationale for its use, as animal and in vitro studies have shown 1-testosterone to have anabolic effects comparable to testosterone, and 1-androsterone is a precursor to this compound. However, direct clinical studies in humans investigating the efficacy and safety of 1-androsterone for bodybuilding outcomes are extremely limited or nonexistent. Most evidence comes from indirect pathways or user anecdotes rather than controlled trials. Because of this, while there is a biochemical basis for its use, the strength of evidence is weak, and safety concerns remain, including potential side effects similar to other anabolic steroids such as liver toxicity, hormonal imbalances, and cardiovascular risks. Regulatory bodies in many countries have banned or restricted its sale due to these concerns and the lack of adequate research. In summary, although there is some scientific foundation for the anabolic potential of 1-androsterone, robust human clinical evidence supporting its use in bodybuilding is lacking.
