Isoleucine is one of the three branched-chain amino acids (BCAAs), along with leucine and valine, and is commonly included in supplements marketed toward bodybuilders and athletes. The rationale for its use centers on the role of BCAAs in muscle protein synthesis and energy production during exercise. While leucine is the primary BCAA implicated in stimulating muscle protein synthesis through activation of the mTOR pathway, isoleucine is thought to contribute to energy regulation and glucose uptake in muscle tissue. Some studies suggest that BCAA supplementation, including isoleucine, may help reduce muscle soreness and support muscle recovery following intense exercise. However, the evidence supporting isoleucine alone as a significant aid for muscle growth or bodybuilding is limited. Most available research examines the effects of BCAAs as a group rather than isoleucine in isolation. Reviews and meta-analyses have concluded that while BCAAs may have modest benefits for muscle recovery, their impact on muscle hypertrophy is inconclusive, especially when adequate protein intake is already present in the diet. Therefore, while isoleucine is scientifically justified as part of BCAA supplementation for athletic recovery, the strength of evidence specifically for bodybuilding benefits is moderate at best.
