Leucine alpha-ketoglutarate (Leu-AKG) is a compound sometimes found in bodybuilding supplements, combining the branched-chain amino acid leucine with alpha-ketoglutarate, an intermediate in the Krebs cycle. The rationale for its use in bodybuilding is based on the individually established roles of leucine and alpha-ketoglutarate. Leucine is well-documented to stimulate muscle protein synthesis via activation of the mTOR pathway, and is a key amino acid for muscle growth and repair. Alpha-ketoglutarate has been investigated for its potential to support energy production and nitrogen balance, which could theoretically support muscle endurance and recovery. However, direct scientific evidence specifically supporting the efficacy of leucine alpha-ketoglutarate as a combined supplement for muscle building is limited. Most available studies have focused on either leucine or alpha-ketoglutarate alone, rather than the combination. Some animal and in vitro studies suggest possible synergistic effects, but human research is scarce. The evidence base for Leu-AKG is thus weak, with little direct clinical trial data in athletes or bodybuilders demonstrating superior muscle growth, strength gains, or recovery compared to standard leucine supplementation. As such, while there is a plausible scientific rationale for its use, the direct evidence remains limited, warranting a moderate evidence rating.
