Rating (out of 5): 3

Testolone (RAD140) is a selective androgen receptor modulator (SARM) that has gained popularity in bodybuilding and athletic circles for its purported muscle-building properties. Scientific studies, primarily preclinical and early-phase human trials, suggest that Testolone can increase lean muscle mass and strength by selectively stimulating androgen receptors in muscle and bone tissue while minimizing the typical androgenic (e.g., prostate and hair) side effects associated with anabolic steroids. Animal studies have demonstrated significant anabolic effects with reduced off-target toxicity. Limited human data—mostly from small phase I/II trials in oncology settings—have shown some increases in lean body mass and tolerability, but robust, long-term, placebo-controlled studies in healthy or athletic populations are lacking.

Despite its popularity, Testolone is not approved by the FDA for any use, including bodybuilding, and its safety profile remains incompletely characterized, with concerns about potential liver toxicity, hormonal suppression, and other adverse effects. The World Anti-Doping Agency (WADA) prohibits SARMs, including Testolone, in competitive sports. In summary, while there is a scientific rationale and some early evidence supporting Testolone’s anabolic effects, the overall strength of evidence for its efficacy and safety in bodybuilding is moderate and incomplete, meriting a rating of 3 out of 5.
