Vitamin B6 (Pyridoxine HCl) has been used traditionally to support or treat carpal tunnel syndrome (CTS), especially in the latter half of the 20th century. This practice originated after early anecdotal reports and small, uncontrolled studies in the 1970s and 1980s suggested that B6 supplementation could alleviate symptoms of CTS. These reports hypothesized that B6 deficiency might contribute to nerve compression or dysfunction, and that supplementation could help restore normal nerve function.

However, subsequent scientific research has failed to provide robust evidence supporting the efficacy of Vitamin B6 for CTS in the general population. Most controlled studies and systematic reviews have found little or no benefit of B6 supplementation over placebo, except in cases where a clear deficiency is present. The American Academy of Orthopaedic Surgeons and other major medical organizations do not recommend Vitamin B6 for CTS unless there is documented deficiency.

Despite the lack of strong scientific validation, the use of B6 for CTS persists in some alternative and complementary medicine circles, largely due to tradition and the relative safety of the vitamin at recommended doses. In summary, the practice is justified primarily by traditional use rather than solid scientific evidence.
