Synopsis

Source of validity: Scientific
Rating (out of 5): 2

Ginkgolide B is a terpene lactone found in the leaves of the Ginkgo biloba tree. Extracts of Ginkgo biloba, standardized to contain ginkgolides (including Ginkgolide B) and flavonoids, have been traditionally used to support cognitive function and cerebral circulation. Scientific investigations have explored the pharmacological properties of Ginkgolide B, noting its role as a platelet-activating factor (PAF) antagonist, which can improve microcirculation and reduce blood viscosity. Several clinical studies and meta-analyses have evaluated Ginkgo biloba extracts (not Ginkgolide B alone) in the context of cognitive impairment and peripheral vascular disease, with some suggesting modest improvement in symptoms such as memory loss or intermittent claudication.

However, the evidence supporting a direct and robust effect of Ginkgolide B (or Ginkgo biloba extracts) on cerebral circulation or cognitive enhancement is mixed. Some studies report statistically significant benefits, while others find little to no effect compared to placebo. Most trials emphasize that the observed effects are generally mild, and methodological limitations (such as small sample sizes and short durations) are common. Thus, while there is scientific rationale and some clinical data supporting the use of Ginkgolide B-containing extracts for brain circulation, the strength of evidence is moderate at best, justifying a rating of 2 out of 5.
