Dodeca-2E,4E,8Z,10Z-tetraenoic acid isobutylamide is one of the principal alkamides found in Echinacea species, particularly Echinacea angustifolia and Echinacea purpurea. Echinacea preparations have a long history of traditional use, especially among Native American populations, for the prevention and treatment of colds and other upper respiratory tract infections. The use of Echinacea became popular in Western herbal medicine in the late 19th and early 20th centuries, primarily for its supposed immune-boosting properties. The alkamides, including dodeca-2E,4E,8Z,10Z-tetraenoic acid isobutylamide, are believed to be among the active components responsible for immunomodulatory effects observed in in vitro and some in vivo studies. However, clinical evidence supporting the efficacy of Echinacea (and its alkamides) for cold prevention is mixed and generally weak. Systematic reviews and meta-analyses of randomized controlled trials have found only modest or no statistically significant benefits in reducing the incidence or duration of colds. The tradition of use is well-established, but scientific validation remains limited, with a low to moderate level of evidence for effectiveness.
