Glycyrrhizic acid, a major active compound in licorice root (Glycyrrhiza glabra), has been investigated for its potential to support or treat colitis, primarily in preclinical studies. The scientific rationale is based on glycyrrhizic acid's anti-inflammatory, immunomodulatory, and antioxidant properties. In animal models of experimental colitis (such as DSS- or TNBS-induced colitis in mice and rats), glycyrrhizic acid administration has been shown to reduce disease severity, attenuate the production of pro-inflammatory cytokines (e.g., TNF-α, IL-1β, IL-6), and decrease histopathological markers of inflammation. Mechanistic studies suggest that glycyrrhizic acid may inhibit NF-κB signaling and modulate macrophage activation, both of which are relevant to the pathophysiology of colitis. However, robust clinical evidence in humans is lacking. Most data come from in vitro or animal studies, with only limited clinical investigations or case reports addressing its efficacy or safety in patients with ulcerative colitis or Crohn’s disease. Traditional use of licorice in gastrointestinal ailments exists, but its application specifically for colitis is not well-documented in historical texts. Overall, while preclinical data provide a promising foundation for glycyrrhizic acid’s potential in colitis, the current scientific evidence is limited and insufficient to warrant strong recommendations for its use in clinical practice without further human studies.
