Glucofrangulin is an anthraquinone glycoside derived from the bark of Rhamnus frangula (commonly known as buckthorn). It is a well-known constituent in many traditional herbal remedies used as laxatives. The laxative effects of glucofrangulin are primarily due to its ability to stimulate peristalsis in the colon and increase water and electrolyte secretion into the intestinal lumen, which facilitates bowel movements and relieves constipation.
There is scientific evidence supporting the efficacy of glucofrangulin and related anthraquinone derivatives for the short-term treatment of constipation in adults. Both the European Medicines Agency (EMA) and the German Commission E recognize Frangula bark preparations (which contain glucofrangulin) as effective for occasional constipation. Several pharmacological studies have demonstrated that anthraquinones exert their effect by being metabolized by gut bacteria into active forms, which then act on the intestinal mucosa to promote laxation. However, the evidence is primarily based on traditional use, some clinical studies, and well-established pharmacological mechanisms rather than large-scale randomized controlled trials specifically evaluating glucofrangulin alone.
While glucofrangulin-containing extracts are effective, they are generally recommended for short-term use only due to potential side effects such as abdominal cramps, dependence, and electrolyte imbalance with prolonged use. Overall, the scientific evidence justifies its use for occasional constipation, but its safety profile necessitates caution and restricts long-term use.
