Bifidobacterium animalis, particularly the subspecies B. animalis subsp. lactis, has been studied for its effects on childhood constipation. Several randomized controlled trials (RCTs) have investigated its efficacy. For instance, a 2010 RCT by Guerra et al. showed that supplementation with B. animalis subsp. lactis DN-173 010 improved stool frequency and consistency in constipated children compared to placebo. Other studies, such as a 2017 meta-analysis by Wojtyniak and Szajewska, found a modest but statistically significant improvement in stool frequency with B. animalis supplementation. However, results across studies are not entirely consistent, with some trials failing to show significant clinical benefit.

The proposed mechanism involves modulation of the gut microbiota composition, production of short-chain fatty acids that promote bowel motility, and improved intestinal barrier function. While traditional use of probiotics for constipation is limited, the use of Bifidobacterium animalis is primarily supported by modern clinical research. Overall, the evidence is moderate (rated 3/5): there is reproducible, though not universally strong, scientific validation for the use of B. animalis in relieving constipation in children, especially for increasing stool frequency. Major pediatric and gastroenterological guidelines recognize that certain probiotics, including B. animalis, may be helpful as adjuncts but are not a substitute for standard therapies.
