Vasicinone is an alkaloid isolated from the plant Adhatoda vasica (also known as Malabar nut or Vasaka), which has a long history of use in traditional Ayurvedic and Unani medicine for the treatment of respiratory ailments, including cough. The use of Adhatoda vasica preparations, which contain both vasicine and vasicinone, is primarily based on traditional knowledge and empirical evidence accumulated over centuries. These preparations are believed to act as expectorants and bronchodilators, providing relief from cough, particularly dry cough.

Limited preclinical studies on vasicinone and its parent plant have shown bronchodilatory activity and some potential to improve mucociliary clearance in animal models. However, direct scientific clinical evidence supporting the efficacy and safety of purified vasicinone for dry cough in humans is lacking. Most of the available evidence is derived from traditional uses and pharmacological investigations in vitro or in animals. Therefore, while its use is well-established in traditional medicine, the scientific validation for vasicinone as a treatment for dry cough remains limited, and high-quality clinical trials are needed.
