Uridine is a nucleoside that plays a critical role in various biological processes, including RNA synthesis and phospholipid metabolism. Its use in supporting the dopamine system has some basis in scientific research, primarily from preclinical studies and a limited number of clinical observations. Preclinical studies in rodents have shown that uridine, especially when combined with other compounds like choline and DHA, can enhance the synthesis and release of dopamine in the brain. This is thought to occur via uridine’s role in promoting membrane phospholipid synthesis, which may facilitate synaptic function and neurotransmitter release, including dopamine. Additionally, uridine supplementation has been investigated for its potential to improve symptoms in mood disorders such as depression and bipolar disorder, conditions that are often associated with dysregulation of dopamine pathways. Some small human studies and case reports suggest potential mood-enhancing effects, but robust, large-scale clinical trials are lacking. Despite these promising leads, the overall evidence supporting uridine's effects on the dopamine system in humans remains limited. Most of the data come from animal studies, and translation to clinical practice is not yet well substantiated. Therefore, while there is a scientific rationale and some supporting evidence, the strength and quality of evidence are currently modest, justifying a mid-to-low evidence rating.
