Synopsis

Source of validity: Traditional
Rating (out of 5): 3

Agnuside is one of the primary active iridoid glycosides found in the fruit of Vitex agnus-castus (chaste tree). Traditionally, Vitex has been used for centuries in herbal medicine, particularly in European and Mediterranean regions, to support female reproductive health. Its primary historical uses relate to the regulation of menstrual cycles, management of premenstrual syndrome (PMS), and relief of mild menopausal symptoms. The "Estrogen body system" is not a common clinical term, but the traditional use of Vitex preparations—containing agnuside—relates to the modulation of hormones involved in the menstrual cycle, particularly by acting on the hypothalamic-pituitary axis to influence prolactin secretion.

While there is some preliminary scientific evidence that agnuside and related compounds may affect hormone balance, the direct impact on estrogen levels is not well established. Most available clinical trials and pharmacological studies have examined whole Vitex extracts, not isolated agnuside, and suggest a dopaminergic effect leading to reduced prolactin rather than direct estrogenic activity. As such, the justification for agnuside supporting the "Estrogen body system" primarily originates from traditional usage, with a moderate (3/5) evidence rating due to the lack of robust clinical trials specifically for agnuside and its direct effects on estrogen.
