Calcium d-Glucarate is a calcium salt of D-glucaric acid, which is found naturally in fruits and vegetables. Its use in supporting estrogen metabolism primarily stems from research on its ability to inhibit the enzyme beta-glucuronidase. This enzyme can deconjugate estrogens in the gut, potentially allowing them to be reabsorbed into circulation rather than excreted. By inhibiting beta-glucuronidase, calcium d-glucarate may promote the excretion of excess estrogens and other toxins. Several animal studies and limited human trials have explored this mechanism, suggesting a potential role in supporting healthy estrogen metabolism and even reducing risk markers associated with hormone-driven cancers. However, most of the direct evidence for its effects on estrogen levels and metabolism comes from animal studies or in vitro work; robust, large-scale clinical trials in humans are lacking. While the mechanistic rationale is plausible and some early-stage evidence exists, the overall quality and amount of clinical data is limited. Thus, although there is scientific interest and some supportive findings, the evidence base does not yet justify a strong recommendation for calcium d-glucarate as a proven supplement for estrogen balance in humans.
