D-glucarate, often used in the form of calcium D-glucarate, is a supplement promoted for its potential role in supporting estrogen metabolism and detoxification. The scientific rationale centers on its ability to inhibit the enzyme beta-glucuronidase in the gut. This enzyme can deconjugate (reactivate) estrogens that the body has already prepared for excretion, potentially allowing them to be reabsorbed and recirculated. By inhibiting beta-glucuronidase, D-glucarate may promote the elimination of excess estrogens and estrogenic metabolites, thus theoretically supporting estrogen balance.
Preclinical studies in animals have shown that D-glucarate supplementation can reduce the activity of beta-glucuronidase and lower the incidence of mammary tumors induced by carcinogens, possibly due to reduced estrogen recirculation. Human data are limited and largely indirect; some small clinical studies have shown changes in markers of estrogen metabolism, but there is no robust evidence from large clinical trials demonstrating clear benefits for hormone balance or estrogen-related conditions. As such, while the mechanism is plausible and there is some supporting animal and in vitro data, the overall level of scientific validation is modest.
In summary, D-glucarate is used based on a mechanistic rationale and supportive preclinical data, but strong clinical evidence in humans is lacking at this time.
