Potassium glucarate is a salt form of glucaric acid, which is naturally found in some fruits and vegetables. The interest in potassium glucarate for estrogen-related support is primarily based on its role as a precursor to D-glucarate, which in the body can be converted to D-glucaro-1,4-lactone. This compound is known to inhibit the enzyme beta-glucuronidase.

Beta-glucuronidase plays a role in the enterohepatic recirculation of estrogens. By inhibiting this enzyme, potassium glucarate may limit the reabsorption of estrogen in the gut, thereby promoting the excretion of estrogen and its metabolites. This mechanism is the scientific rationale for using potassium glucarate to "support estrogen metabolism" or "detoxification". Some animal studies and in vitro research have shown that D-glucarate supplementation can decrease beta-glucuronidase activity and reduce levels of circulating estrogens, which has led to interest in its potential protective effects against estrogen-sensitive cancers.

However, human clinical evidence is limited and mostly indirect. While the biochemical mechanism is plausible and supported by preclinical studies, well-controlled clinical trials demonstrating clear benefits for estrogen-related health in humans are lacking. Thus, while there is a scientific basis for its use, the strength of the evidence is low to moderate, warranting a rating of 2 out of 5.
