β-nicotinamide mononucleotide (NMN) is a precursor to nicotinamide adenine dinucleotide (NAD+), a molecule crucial for cellular energy metabolism, DNA repair, and mitochondrial function. In the context of Fatty Liver Disease—particularly non-alcoholic fatty liver disease (NAFLD)—scientific interest has grown regarding NMN’s potential. Preclinical animal studies have shown that NMN supplementation increases hepatic NAD+ levels, enhances mitochondrial function, and improves insulin sensitivity, all of which are impaired in fatty liver conditions. For instance, some mouse models of NAFLD demonstrate reduced hepatic steatosis and inflammation after NMN administration, likely due to improved lipid metabolism and reduced oxidative stress.
However, human clinical evidence remains limited. As of mid-2024, there are only a handful of small-scale human trials investigating NMN’s effects on metabolic health, and even fewer directly targeting fatty liver disease. Most of these trials focus on endpoints like insulin sensitivity or general markers of liver function, rather than direct measurement of hepatic fat content or liver histology. Thus, while the mechanistic rationale for NMN use in fatty liver is strong based on animal studies and basic science, robust clinical validation in humans is still lacking. Overall, scientific evidence is promising but preliminary, justifying a moderate evidence rating and underscoring the need for larger, well-controlled human studies.
