Alpha Methyl Tetradecylthioacetic Acid (also known as Tetradecylthioacetic acid or TTA) has been investigated in preclinical and limited clinical studies as a potential treatment for Fatty Liver Disease (hepatic steatosis). TTA is a synthetic fatty acid analogue that acts primarily as a pan-agonist of peroxisome proliferator-activated receptors (PPARs), particularly PPAR-α, which plays a role in lipid metabolism and fatty acid oxidation.

Animal studies have demonstrated that TTA can reduce hepatic lipid accumulation, improve insulin sensitivity, and decrease inflammation associated with non-alcoholic fatty liver disease (NAFLD). In rodent models, TTA supplementation led to reduced liver fat content and improved liver function markers.

Human data, however, are quite limited. A small pilot study in obese humans with type 2 diabetes (N = 10) found that TTA supplementation for 4 weeks lowered plasma triglycerides and improved some markers of lipid metabolism, but there was no significant change in liver fat content as measured by imaging. Other early-phase trials have focused on lipid profiles and markers of inflammation rather than direct measures of hepatic steatosis.

In summary, while there is experimental and early clinical evidence supporting a mechanistic role for TTA in lipid metabolism and fatty liver, the current clinical evidence is insufficient for strong recommendations. Its use is primarily supported by preclinical studies and limited clinical trials, and further well-controlled human studies are needed to establish efficacy and safety for fatty liver disease.
