Lupulinum refers to the glandular powder from the strobiles (cones) of Humulus lupulus (hops). The use of lupulinum to support the GABA system is primarily based on traditional herbal medicine rather than on strong scientific validation. Historically, hops have been used as a mild sedative and sleep aid, with documented usage in European herbal traditions to calm restlessness and ease insomnia. The rationale for its calming effects is attributed to bioactive compounds such as humulone, lupulone, and xanthohumol, which are thought to interact with the central nervous system.
While there is some preclinical evidence suggesting that extracts from hops may modulate GABAergic activity (the neurotransmitter system associated with relaxation and sedation), direct clinical studies on lupulinum’s efficacy and its mechanism of action are limited. Animal and in vitro studies have indicated that certain hop constituents may bind to GABAA receptors, producing sedative effects, but robust human clinical trials are lacking.
