Agnuside is a primary active constituent found in the fruit of Vitex agnus-castus (chaste tree). Traditionally, this plant has been used for centuries in European herbal medicine, particularly to support hormone balance and reproductive health in women. Its use is mainly associated with the regulation of menstrual cycles, relief from premenstrual syndrome (PMS), and management of certain symptoms related to menopause. The rationale for its effect on the glandular (endocrine) system is based on historical observations that suggest Vitex agnus-castus can influence pituitary gland function, especially the secretion of prolactin, thereby indirectly affecting other hormones. However, most of the supporting evidence comes from the whole plant extract rather than isolated agnuside. While some preliminary scientific studies have shown that Vitex extracts may modulate hormone levels, the direct role of agnuside itself in supporting the glandular system is not well established in clinical research. Nonetheless, due to the plant’s long-standing traditional use for hormonal and glandular imbalances, the use of agnuside-containing preparations is considered to be justified primarily by tradition, with moderate but not robust evidence.
