8-Prenylnaringenin (8-PN) is a phytoestrogen, most notably found in hops (Humulus lupulus), and is recognized as one of the most potent plant-derived estrogens. Its use for supporting or treating hot flashes, a common symptom of menopause, has scientific backing primarily due to its estrogenic activity. Several in vitro and animal studies have demonstrated that 8-PN can bind to estrogen receptors, particularly ERα, mimicking the effects of endogenous estrogens. Human clinical studies are limited but promising: small randomized controlled trials and open-label studies have shown that hop extracts standardized for 8-PN content may reduce the frequency and severity of hot flashes in peri- and postmenopausal women. For example, a 2013 double-blind, placebo-controlled trial reported a significant reduction in hot flash scores with hop extract compared to placebo over 12 weeks. However, the number of high-quality, large-scale human clinical trials is still low, and most studies use hop extract rather than pure 8-PN, making it difficult to attribute effects solely to 8-PN. The current level of evidence supports the potential efficacy of 8-PN for hot flashes, but further rigorous research is needed to establish optimal dosing, safety, and long-term effects.
